No, don't press "Define". Just enter wildcards, e.g.
*.*
or
*.txt *.htm *.html
then click on "OK". Then click on "+" and choose "tc" -> "Compare as text".

I can't find this option in 10.52.
I think the best solution is to add checkbox "Ignore new lines".
