Everytime I try and Change the Button Bar, or Change an item in the Custom Columns Dialog (add/remove a column item) -- TC locks up for 30 seconds on more and the dialog in question changes title to "Not Responding" :: Windows offers me the option of closing the App, but if I wait a little longer then it does recover.
This is occuring on a School Computer
OS: Vista Business 32bit.
TC: 7.55a, Running on a FlashDrive.
AV: Symantec Anti-Virus -- I added an Exclusion to Live Scanning for the USB Drive.
I tried Starting TC with an empty wincmd.ini file...
I have tried starting it from a Command Prompt that has Admin Rights, and a Command Prompt that doesn't have Admin Rights. As well as by just directly launching TC by clicking on its EXE file.
In all instances the freezing continues whenever I try and modify the button bar or a Custom Column.

Sounds indeed like a virus scanner - try to exclude the wincmd.ini and default.bar from being scanned.
If that doesn't help, try using "process monitor" to find out which function takes that long.
