Yes, is it possible to make Total Commander close with long-press in Android 16?

You mean long press on the back button? Unfortunately this is no longer possible.
It's because of the new "predictive back" feature. You can find out more here:

Even when opting out from predictive back, the back button long press is no longer sent to the app.
