For several of the buttons It would be a multi-line tooltip - I think that would get annoying if it popped up every time you happen to hover a Fn button.Or perhaps a tooltip for all modifiers could be shown when hovered even if nothing is pressed?

It's not enough. In this case you can't see that any of Alt, Ctrl or Shift keys stuck in a pressed position.

You can change system settings to show underlined hotkey letters in menu only after pressing Alt or to always show hotkeys. In the first case you can see when the Alt key is stuck.
Do you mean that it is Windows Explorer's job to be responsible for revealing physically stuck Alt? For me it's just a nice side effect.
To be serious, all TC features should be available from keyboard WITH NO mouse or other pointing device present. See my other message about drive selection in Alt+F7 options:

That could be obtained the same way as it is for the Button bar - here you have to focus the Button bar with Alt+Shift+F11 before you can see the tooltips for the buttons without using the mouse. So you want me to navigate from button to button and pressing Alt many times rather than just showing all functions immediately after pressing modifier key. It's completely user unfriendly.

I imagined that you hold down Alt and move between the Function keys - with either the mouse - or the arrow keys once the function key bar has focus, if you don't want to use the mouse.So you want me to navigate from button to button and pressing Alt many times rather than just showing all functions
I am not against a solution where the text on the buttons changes when the modifier keys are pressed, but that will need a lot of new language strings that fits the buttons..

Using Nirsoft's tool KeyboardStateView you can easily see that all Alt Ctlr Shift keys can be queried: That's why I'm in favor of the following:

As I said: Each user enters their own text there individually. If they do not enter any, the standard text is displayed.
One line each for a combination and then the texts with "|" as separator. (space/notext for standard text and - for 'no text').
