Different icon for folders containing only files, and folders contains subfolders (and files)
Is there a way to change a folder icon if that folder contains another folder(s), possibly in addition to files? I need to quickly identify folders that contain other folders.

No, but you can colorize the folders that have subfolders by using content plugins (tc and DirSizeCalc) with the function "Define color by filetype"
